However, the progression-free survival (PFS) for first-line and second-line treatments remains poorly characterized in hepatopancreatobiliary cancers. Furthermore, some patients are willing to receive treatment but maybe not suitable for intravenous chemotherapy. The OS for supportive care only is also important for designing a first-line trial focused on chemotherapy-ineligible patients. As some mutations are increasingly recognized as a potential druggable target, establishing benchmarks for first-line and second-line PFS\u2014which is the FDA-endorsed surrogate endpoint for accelerated approval in phase II trials\u2014is crucial.
